Inenzymology, a2-ethylmalate synthase (EC2.3.3.6) is anenzyme thatcatalyzes thechemical reaction

acetyl-CoA + H2O + 2-oxobutanoate{\displaystyle \rightleftharpoons } (R)-2-ethylmalate + CoA

The 3substrates of this enzyme areacetyl-CoA,H2O, and2-oxobutanoate, whereas its twoproducts are(R)-2-ethylmalate andCoA.

This enzyme belongs to the family oftransferases, specifically thoseacyltransferases that convert acyl groups into alkyl groups on transfer. Thesystematic name of this enzyme class isacetyl-CoA:2-oxobutanoate C-acetyltransferase (thioester-hydrolysing, carboxymethyl-forming). Other names in common use include(R)-2-ethylmalate 2-oxobutanoyl-lyase (CoA-acetylating),2-ethylmalate-3-hydroxybutanedioate synthase,propylmalate synthase, andpropylmalic synthase. This enzyme participates inpyruvate metabolism.
