Tolerance and Tachyphylaxis to Head Twitches Induced by the 5-HT2A Agonist 25CN-NBOH in Mice

Abstract:The serotonin (5-HT) 2A receptor is the primary molecular target of serotonergic hallucinogens, which trigger large-scale perturbations of the cortex. Our understanding of how 5-HT2A activation may cause the effects of hallucinogens has been hampered by the receptor unselectivity of most of the drugs of this class. Here we used 25CN-NBOH (N-(2-hydroxybenzyl)-2,5-dimethoxy-4-cyanophenylethylamine), a newly developed selective 5-HT2A agonist, and tested it with regard to the head-twitch-response (HTR) model of 5… Show more

“…We chose the timing of the LSD and 25CN-NBOH administration such that the onset of the effects expressed as head twitch responses would overlap with the gambling task. Based on earlier reports, the increase in head twitches is apparent within minutes after administration, peaking at approximately 5-7 min after administration, and remaining enhanced for more than 30 min, the time course of which therefore overlapped with the entire gambling task session(Halberstadt and Geyer, 2013;Buchborn et al, 2018). As similar head twitch responses are not induced by amphetamine, its administration schedule was based on a prior report of its effects in the Iowa Gambling Task(van Enkhuizen et al, 2013).…”

“…The LSD doses were chosen to cover the range of the most commonly used doses in the literature(Stasik and Kidwell, 1969;Halberstadt and Geyer, 2013;Kyzar et al, 2016;Alper et al, 2018). The dose of 25CN-NBOH was chosen based on its ability to prominently induce head twitch responses in earlier publications(Buchborn et al, 2018;Halberstadt et al, 2019).…”

“…The 25CN-NBOH hydrochloride (a kind gift from Jesper L. Kristensen, University of Copenhagen) was dissolved in isotonic saline and applied subcutaneously (<10 mL/kg). The dose used (1.5 mg/kg) was found optimal for a 5-HT 2A R-specific effect, as determined by a dose-response curve for 25CN-NBOH-induced head twitches ( Buchborn et al, 2018 ). Experiments were performed under general anaesthesia using isoflurane (IsoFlo; Zoetis, UK) delivered in 100% oxygen, as detailed below.…”

“…For AD, FDA-approved treatments focus on cholinesterase (donezepil) [ 59 ], and N-Methyl-D-Aspartic Acid (NMDA) receptors(memantine) [ 60 ]. Many researches on reducing Aβ plaque-secretase modulators (BACE1 [ 60 , 61 , 62 , 63 ]), hyperphosphorylation tau protein [ 64 , 65 ], inflammation [ 66 , 67 ], other oxidative molecules (metal ion [ 68 ], peroxide [ 69 ]) have been studied. For PD, α-syn abnormal aggregation causing impaired motor function with slow movements and tremor [ 70 ], studies mainly concentrate on dopamine [ 71 ].…”
