A novel Zika virus mouse model reveals strain specific differences in virus pathogenesis and host inflammatory immune responses
Abstract:Zika virus (ZIKV) is a mosquito borne flavivirus, which was a neglected tropical pathogen until it emerged and spread across the Pacific Area and the Americas, causing large human outbreaks associated with fetal abnormalities and neurological disease in adults. The factors that contributed to the emergence, spread and change in pathogenesis of ZIKV are not understood. We previously reported that ZIKV evades cellular antiviral responses by targeting STAT2 for degradation in human cells. In this study, we demons… Show more

“…ZIKV-injected pups showed normal performance in reflex evaluation tests carried out early after infection. These results are in contrast with a previous report of impaired performance in the righting reflex induced by an African ZIKV strain(34), which is much more aggressive to the murine nervous system than the strain used in the present study(36). However, ZIKV-infected mice showed several motor function impairments throughout development and during adulthood, suggesting that ZIKV infection has a long-lasting effect on motor performances.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD).Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD.The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC).Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…In this study, using biorthogonal system, we found that ZIKV infection suppressed host de novo translation, and reduce the in the expression of STAT2, while ZIKV NS5 did not suppressed host de novo translation. In line with previous reports [ 22 , 23 , 54 , 55 ], we found that exogenous murine STAT2 was refractory to ZIKV infection-induced degradation ( Figure 7 (a)), which is probably due to the failure of NS5 recognition of murine STAT2. Supporting this point, we found that overexpression of ZIKV NS5 did not affect murine STAT2 protein level ( Figure 7 (b)).…”

“…1A and B ). We found that strain H/PF/2013 was the most virulent, causing 80% lethality, whereas strain FSS13025 caused modest weight loss in some mice and only 20% lethality, and strain PRVABC59 caused no weight loss or lethality, altogether consistent with prior studies reporting the relative virulence of these strains in Ifnar1 −/− mice of various ages and inoculation routes ( 25 , 26 , 28 ). We chose to use strain FSS13025 for further experiments to achieve robust maternal infection without severe maternal morbidity and because of its use in studies from other groups evaluating the role of IFN signaling in congenital ZIKV infection ( 29 ).…”
