Chemical Reprogramming of Human Somatic Cells to Pluripotent Stem Cells.Jingyang Guan,GuanWang,JinlinWang,Zhengyuan Zhang,Yao Fu,Lin Cheng,Gaofan Meng,Yulin Lyu,Jialiang Zhu,Yanqin Li,YangluWang,Shijia Liuyang,Bei Liu,Zirun Yang,Huanjing He,Xinxing Zhong,Qijing Chen,Xu Zhang,Shicheng Sun,Weifeng Lai,Yan Shi,Lulu Liu,LipengWang,Cheng Li,Shichun Lu &Hongkui Deng -forthcoming -Nature:1–7.detailsCellular reprogramming can manipulate the identity of cells to generate the desired cell types1– 3. The use of cell intrinsic components, including oocyte cytoplasm and transcription factors, can enforce somatic cell reprogramming to pluripotent stem cells4– 7. By contrast, chemical stimulation by exposure to small molecules offers an alternative approach that can manipulate cell fate in a simple and highly controllable manner8– 10. However, human somatic cells are refractory to chemical stimulation owing to their stable epigenome2,11,12 and reduced plasticity13,14; it (...) is therefore challenging to induce human pluripotent stem cells by chemical reprogramming. Here we demonstrate, by creating an intermediate plastic state, the chemical reprogramming of human somatic cells to human chemically induced pluripotent stem cells that exhibit key features of embryonic stem cells. The whole chemical reprogramming trajectory analysis delineated the induction of the intermediate plastic state at the early stage, during which chemical-induced dedifferentiation occurred, and this process was similar to the dedifferentiation process that occurs in axolotl limb regeneration. Moreover, we identified the JNK pathway as a major barrier to chemical reprogramming, the inhibition of which was indispensable for inducing cell plasticity and a regeneration-like program by suppressing pro-inflammatory pathways. Our chemical approach provides a platform for the generation and application of human pluripotent stem cells in biomedicine. This study lays foundations for developing regenerative therapeutic strategies that use well-defined chemicals to change cell fates in humans. (shrink)

Bezier Smooth Support Vector Classification.Q. Wu &EnWang -2015 -Journal of Computational Information Systems 11 (12).detailsA new smooth method for solving the support vector machine classification (SVC) is presented. Since the objective function of the unconstrained SVC is non-smooth, we apply the smooth technique and replace the SVC function with Bézier function and get a class of Bézier smooth support vector machines (BSSVM). The fast Newton-Armijo algorithm is used to solve the BSSVM. Theoretical analysis and numerical results illustrate that this smooth SVM model improves in efficiency and accuracy compared with other smooth methods.

A Davidsonian Response to Radical Scepticism.JuWang -2015 -Logos and Episteme 6 (1):95-111.detailsIn this paper, I attempt to show how Davidson’s anti-sceptical argument can respond to the closureRK-based radical scepticism. My approach will focus on the closureRK principle rather than the possibility that our beliefs could be massively wrong. I first review Davidson’s principle of charity and the triangulation argument, and then I extract his theory on content of a belief. According to this theory, content of a belief is determined by its typical cause and other relevant beliefs. With this constraint on (...) content, I argue that doubt must be local. Furthermore, since one cannot rationally believe that one’s commitment to the cause of beliefs could be false, our commitment to the denial of a sceptical hypothesis is not a knowledge-apt belief. Therefore, the closureRK principle is not applicable to rational evaluations of this commitment. As a result, the closureRK-based sceptical argument fails while the closureRK principle remains. Aesthetics, Morality, and the Modern Community:Wang Guowei, Cai Yuanpei, and Lu Xun.BanWang -2020 -Critical Inquiry 46 (3):496-514.detailsIn Mao’s era, China’s policy makers and intellectuals viewed aesthetic experience and thought as handmaidens in the service of the political order. As China opened up and engaged more intensely with modern traditions of the West, aesthetic thinkers such as Li Zehou critiqued the subordinated role of aesthetics and reasserted notions of aesthetic autonomy and liberal humanism, calling for the separation of arts and literature from political, social, and moral concerns. This truncated aesthetic view stems from a modernist version of (...) Western aesthetics that favors sense and sensibility as the sole sources of aesthetic value—a view that considerably diminishes the horizon of concerns inherent in traditional and modern Chinese aesthetic thoughts. This article seeks to recapture a repoliticized aesthetics by considering the ways early twentieth-century Chinese thinkers linked aesthetic thoughts to moral and political considerations in the reconstruction of China as a modern nation-state. Instead of transcendent, pure aesthetics over and above political processes and social experience, prominent aesthetic thinkers likeWang Guowei, Cai Yuanpei, and Lu Xun invoked traditional Chinese aesthetic legacies and reconsidered intimate and necessary ties between aesthetic categories and broader concerns about cultural crises, moral reform, and nation building. Association of Stress-Related Factors With Anxiety Among Chinese Pregnant Participants in an Online Crisis Intervention During COVID-19 Epidemic.Fangfang Shangguan,RuoxiWang,Xiao Quan,Chenhao Zhou,Chen Zhang,Wei Qian,Yongjie Zhou,Zhengkui Liu &Xiang Yang Zhang -2021 -Frontiers in Psychology 12.detailsBackground: Previous systematic review indicated the prevalence of prenatal anxiety as 14–54%. Pregnant women are a high-risk population for COVID-19. However, the prevalence of anxiety symptoms and related factors is unknown in Chinese pregnant women during COVID-19 outbreak. Objective: To investigate the prevalence of anxiety symptoms and the related factors in Chinese pregnant women who were attending crisis intervention during the COVID-19 pandemic. Methods: The data of this cross-sectional study were collected in about 2 months. Data analysis was performed from April to (...) May 2020. Participants completed a set of questionnaires via the Wechat Mini-program before starting the online self-help crisis intervention for COVID-19 epidemic. A total of 2,120 Chinese pregnant women who were attending a self-help crisis intervention participated in this study. A survey was developed to address possible stress-related factors in pregnant women during the COVID-19 outbreak, including demographic, socioeconomic, and pregnancy-related factors, as well as COVID-19 related factors. Generalized Anxiety Disorder-7 scale and the 10-item perceived stress scale were, respectively, employed to measure anxiety and stress-related factors. Results: A total of 21.7% of pregnant women reported at least mild anxiety, and only 82 women reported moderate to severe anxiety. Factors associated with at least mild anxiety included living in Hubei province, nobody providing everyday life support, pelvic pain or vaginal bleeding, and higher perceived stress. Having relatives or neighbors with a diagnosis of COVID-19 was not associated with anxiety. Conclusions and Relevance: Our findings indicate that evaluation and intervention for maternal and infant health are necessary in pregnant women with anxiety during COVID-19 epidemic, especially those with higher perceived stress, less everyday life support, or vaginal bleeding. Interactions among these related medical, social and psychological factors need to be investigated in future studies. (shrink)
Research on transformer vibration monitoring and diagnosis based on Internet of things.Amit Sharma &ZhenzhuoWang -2021 -Journal of Intelligent Systems 30 (1):677-688.detailsA recent advent has been seen in the usage of Internet of things (IoT) for autonomous devices for exchange of data. A large number of transformers are required to distribute the power over a wide area. To ensure the normal operation of transformer, live detection and fault diagnosis methods of power transformers are studied. This article presents an IoT-based approach for condition monitoring and controlling a large number of distribution transformers utilized in a power distribution network. In this article, the (...) vibration analysis method is used to carry out the research. The results show that the accuracy of the improved diagnosis algorithm is 99.01, 100, and 100% for normal, aging, and fault transformers. The system designed in this article can effectively monitor the healthy operation of power transformers in remote and real-time. The safety, stability, and reliability of transformer operation are improved. (shrink)

More for free: A dynamic epistemic framework for conformant planning over transition systems.Yanjun Li,Quan Yu &YanjingWang -2017 -Journal of Logic and Computation 27 (8):2383--2410.details© The Author, 2017. Published by Oxford University Press. All rights reserved. In this article, we introduce a lightweight dynamic epistemic logical framework for automated planning under initial uncertainty. We generalize the standard conformant planning problem in AI in two crucial aspects: first, the planning goal can be any formula expressed in an epistemic propositional dynamic logic ; second, procedural constraints of the desired plan specified by regular expressions can be imposed. We then reduce the problem of generalized conformant planning (...) to the model checking problem of our logic. Although our conformant planning problem is much more general than the standard one with Boolean goals and no procedural constraints, the complexity is still PSPACE-complete which is equally hard as standard conformant planning over explicit transition systems. A pragmatic case against pragmatic theological realism.Wang-yen Lee -2009 -Heythrop Journal 50 (3):479-494.detailsPragmatic theological realism (PTR) urges us to take up the realist aim of theology or the goal of truth although we have good reason to think that the goal can neither be attained nor approximated. Rescher contends that pursuing an unreachable goal can be rational on pragmatic grounds so long as pursuing the unreachable goal yields indirect benefits. I have blocked this attempt at providing a pragmatic justification for the realist aim of PTR on precisely the same pragmatic grounds: since (...) there is a competing alternative to PTR and the alternative can provide whatever indirect benefits PTR can offer while being less risky than it is, prudential reasoning favours the alternative to PTR. This undermines the pragmatic case for the realist aim of theology since the instrumentalist alternative does not aim at the truth. (shrink)
Hitchcock and Sober on Weak Predictivism.Wang-Yen Lee -2012 -Philosophia 40 (3):553-562.detailsAccording to Hitchcock and Sober’s argument from overfitting for weak predictivism, the fact that a theory accurately predicts a portion of its data is evidence that it has been formulated by balancing simplicity and goodness-of-fit rather than overfitting data. The core argument consists of two likelihood inequalities. In this paper I show that there is a surprising accommodation-friendly implication in their argument, and contend that it is beset by a substantial difficulty, namely, there is no good reason to think that (...) their second likelihood inequality is true. Should the No-Miracle Argument Add to Scientific Evidence?Wang-Yen Lee -2014 -Philosophia 42 (4):999-1004.detailsLipton contends that the no-miracle argument is illegitimate, because it fails to adduce new evidence beyond that cited by scientists for their theories. The debate on this issue between Lipton and Psillos has focussed on whether there is a construal of the no-miracle argument in relation to first-order scientific inferences that can yield new evidence. I move away from this focus without taking sides, and argue that the no-miracle argument, on its two popular interpretations, is as legitimate, cogent, and useful (...) an inductive argument for scientific realism as first-order scientific inferences to the best explanation even if it does not add to scientific evidence. Face recognition method based on multi-class classification of smooth support vector machine.Wang En Wu Qing, Liang Bo,Wang Wan &EnWang -2015 -Journal of Computer Applications 35 (s1).detailsA new three-order piecewise function was used to smoothen the model of Support Vector Machine( SVM) and a Third-order Piecewise Smooth SVM( TPSSVM) was proposed. By theory analyzing, approximation accuracy of the smooth function to the plus function is higher than that of the available. When dealing with the multi-class problem, a coding method of multi-class classification based on one-against-rest was proposed. Principal Component Analysis( PCA) was employed to extract the main features of face image set, and multi-class classification of (...) smooth SVM was used for face recognition. The experimental results on ORL and FERET face databases show that the recognition rate of smooth SVM for multi-class classification is better than the traditional identification methods. (shrink)
