Abstract

Methods, devices and systems are disclosed for generating and harvesting skin grafts having improved properties and for ensuring efficient and consistent blister formation and/or reducing patient harm and discomfort. In one aspect, the invention infuses a fluid into skin tissue at the donor site to enhance blister formation. The fluid infusion can occur before or after application of negative pressure—or can be cyclically applied before or after repeated applications of negative pressure.

Description

Claims (20)

What is claimed is:
1. A method for harvesting a skin graft, the method comprising:
applying a blister cutting device to a target region of a subject's skin;
infusing a hypotonic fluid into the target region of the subject's skin;
generating a negative pressure within the blister cutting device to raise at least one skin blister on the target region of the subject's skin; and
cleaving the at least one blister using the blister cutting device.
2. The method ofclaim 1, further comprising warming the hypotonic fluid to a temperature ranging between at least one of: 20° C. and 40° C., 25° C. and 35° C., and 28° C. and 33° C.
3. The method ofclaim 1, further comprising exposing the target region of the subject's skin to at least one of: purified water, deionized water, water having an electrical conductivity less than 11 microsiemens/cm, and water having an electrical conductivity less than about 5 microsiemens/cm.
4. The method ofclaim 1, wherein the hypotonic fluid comprises a fluid having a lower osmotic pressure than interstitial fluid in the skin at the target region.
5. The method ofclaim 1, further comprising exposing the target region of the subject's skin to the hypotonic fluid for a time duration ranging between at least one of about 1 minute and 30 minutes and about 5 minutes and 10 minutes.
6. The method ofclaim 1, further comprising infusing additional hypotonic fluid into the target area of the subject's skin following generation of the negative pressure within the blister cutting device.
7. The method ofclaim 6, further comprising re-applying the negative pressure to the target area of the subject's skin following infusion of the additional hypotonic fluid.
8. The method ofclaim 1, further comprising applying an adhesive substrate to the at least one blister prior to cleavage of the at least blister and harvesting the skin graft by removing the substrate from blister cutting device following the cleavage.
9. The method ofclaim 8, wherein the at least substrate comprises a water resistant adhesive.
10. The method ofclaim 8, further comprising drying the target region prior to applying the adhesive substrate.
11. The method ofclaim 1, wherein the blister cutting device comprises at least one fixed plate and at least one movable cutter plate, each plate having a plurality of holes through which skin blisters can be raised when the plurality of holes in the at least one fixed plate and the plurality of holes in the at least one movable cutter plate are aligned, and further comprising cleaving the at least one blister by moving the at least one movable cutter plate to disrupt alignment of the plurality of holes and separate the at least one blister from the target region of the subject's skin.
12. The method ofclaim 1, further comprising exposing the target region of the subject's skin to at least one of an anesthetic agent and an antibiotic agent.
13. A method for harvesting a skin graft, the method comprising:
applying a device having a body and a sealing member to a target region of a subject's skin, the body and the sealing member being configured to form a sealed chamber over the target region of the subject's skin and the device further comprising at least one fixed plate and at least one movable cutter plate, each plate having a plurality of holes through which skin blisters can be raised when the plurality of holes in the at least one fixed plate and the plurality of holes in the at least one movable cutter plate are aligned;
infusing a hypotonic fluid into the sealed chamber and delivering the hypotonic fluid for assimilation into portions of the subject's skin exposed to the sealed chamber via openings of the plurality of holes in the at least one fixed plate and at least one movable cutter plate;
removing unassimilated fluid from the sealed chamber;
generating a negative pressure within the sealed chamber to raise at least one skin blister through the openings of the plurality of holes; and
actuating the at least one movable cutter plate to cleave the at least one skin blister.
14. The method ofclaim 13, further comprising opening the sealed chamber prior to cleavage of the at least one skin blister and applying an adhesive substrate to the at least one blister formed through the openings of the plurality of holes.
15. The method ofclaim 14, further comprising harvesting the skin graft by removing the substrate from device following the cleavage of the at least one skin blister.
16. The method ofclaim 15, further comprising transferring the skin graft to a recipient site on a patient's skin by applying the substrate having the at least one skin blister to the recipient site.
17. The method ofclaim 13, wherein the device further comprises a fluid heater and further comprising warming the hypotonic fluid via the fluid heater.
18. The method ofclaim 13, further comprising warming the hypotonic fluid to a temperature ranging between at least one of: 20° C. and 40° C., 25° C. and 35° C., and 28° C. and 33° C.
19. The method ofclaim 13, wherein the hypotonic fluid comprises at least one of: purified water, deionized water, water having an electrical conductivity less than 11 microsiemens/cm, water having an electrical conductivity less than about 5 microsiemens/cm, and a fluid having a lower osmotic pressure than interstitial fluid in the skin at the target region.
20. The method ofclaim 13, wherein the device further comprises a reservoir configured to contain the hypotonic fluid and a conduit configured to fluidly couple the reservoir to the sealed chamber, and further comprising delivering the hypotonic fluid from the reservoir to the sealed chamber via the conduit.
