Abstract

PURPOSE:To obtain an enzyme electrode has superior stability in preservation and speeds and facilitates measurement by fixing oxidoreductase on an insoluble metallic complex and by bringing this complex into electric contact with a conductive substance. CONSTITUTION:On a disk 1 obtained by press-forming oxidoreductase, such as glucose oxidase, insoluble metallic complex, such as iron, copper and manganese, constituting a redox system, and conductive substance (graphite) powder, or by forming and sticking the insoluble metallic complex on the compact after compacting the conductive powder, enzyme 4 is fixed to manufacture an enzyme electrode 5. The electrode 5 is fixed in a cylindrical holder 6 with a screw 7 and a platinum plate current collecting body 8 is contacted and connected to a lead wire 9. Then, the density of a substrate (glucose, etc.) is measured by a device which uses a saturation calomel electrode 10 as a reference electrode, a counter electrode 11 and a phosphoric acid buffer solution 12 as an electrolyte and is equipped with a separator 13. The electrode 5 is stable in preservation and its characteristics are maintained for a long period.
