An ampoule and needle assembly comprises an ampoule having a plunger at one end and a closure at the other, and a double-ended hypodermic needle mounted on a needle holder which is guided by the ampoule while communication with the interior of the ampoule is being established. In one form the ampoule body 12 has a piston type plunger 13 and a rubber closure member 20, the latter having a recess 27 extending partly therethrough so as to leave a thin diaphragm 28. The needle is carried in a cylindrical holder 30, an extension 31 of which fits into the recess 27. When the ampoule is fitted into a holder, forward movement of the body 12, relative to the needle, causes the latter to pierce the diaphragm 28. The needle cover 40 is then removed for the injection to be performed. In another type of assembly the needle holder has a flange which fits over the outside of the neck of the ampoule. A suitable type of holder is described with reference to Figs. 3 and 4 (not shown).
