1420916 Analytical filter K D BAGSHAWE 8 Dec 1972 [8 Sept 1971 12 Sept 1972] 41972/71 and 42383/72 Heading B1D [Also in Division Gl] A reaction cell, for use in for example radioimmunoassay, comprises a tubular element 11 having enlarged upper and lower parts 12, 13 and a central region containing a matrix pad 17 lodged on an apertured support 16, the matrix pad comprising a porous absorptive material, for example glass fibre filter material. The pad may contain a predetermined amount of antigen or antibody and a pad of fibrous material may be coated before introduction of the reagent with inert maoromolecular material such as albumin. The cell may contain more than one matrix pad. The cell may be fitted to upper member 14, which has prongs 20 which hold the pads in position when wash liquid is introduced at the upper end of the assembly and to lower member 15, which may contain cellulose wadding or absorbent tissue extending slightly above member 15 to touch the matrix pad through aperture 25 and so aid filtration. In use, for example in the determination of digoxin, preprepared matrix pads containing a mixture of rabbit anti-digoxin serum and donkey anti-rabbit serum are inserted into respective reaction cells. Plasma samples being tested and standard solutions are mixed I-125 labelled digoxin and aliquots of of the mixtures pipetted into respective reaction, cells. The upper tubes 14 are attached, the assemblies incubated, the lower tubes 15 are attached, phosphate albumin buffer is introduced into the upper tubes to wash unreacted I-125-labelled digoxin from the pads and the amount of radioactivity left in the pads is determined.
