Background
Dendrobium officinale is a perennial herb of Dendrobium of Orchidaceae, commonly known as Heijiecao, is a traditional and famous and precious traditional Chinese medicinal material, is mainly distributed in Guangxi, Yunnan, Zhejiang and Fujian places and has the effects of tonifying stomach, promoting the production of body fluid, nourishing yin, clearing heat, improving human immunity, promoting gastrointestinal tract digestion, resisting aging and the like. The product has unique medicinal value and health promotion effect, and can be used as health promotion tonic. At present, the problems of putrefaction, color change, water loss, germination and the like of fresh dendrobium officinale are easily caused in the storage process due to the immature dendrobium officinale preservation technology. Therefore, the picked fresh dendrobium candidum strips are generally fried into spiral dendrobium candidum. Although the fried dendrobium candidum has longer shelf life than the fresh dendrobium candidum, part of nutrient components are lost in the frying process. Therefore, the market has great demand on the preservation method and the preservative for the fresh dendrobium officinale strips.
Ferulic acid is a phenolic acid commonly existing in cell walls of natural plants, and is widely applied to food and medicine industries due to good effects of oxidation resistance, antibiosis, cancer resistance and the like. However, the ferulic acid ester has poor solubility and unstable physicochemical properties, so the application of the ferulic acid ester in food is limited to a certain extent. Researches show that the ferulic acid ester derivative obtained by molecular modification not only embodies and maintains the biological characteristics of ferulic acid, but also has lower toxicity and stronger antibacterial and antioxidant properties. The existing research finds that the ferulic acid hexyl ester has strong bacteriostatic action and antioxidant action in a series of ferulic acid fatty acid esters with different carbon chain lengths, can effectively inhibit the growth of food-borne pathogenic bacteria, and slows down the hydrolysis and rancidity of grease.
The silver carp is popular with consumers due to its fast growth speed, high yield and rich nutrition. However, a huge amount of fish processing byproducts (fishbone, fish skin, fish scale) are formed each year, and the market potential is often neglected. In addition, improper disposal of these fish processing by-products is also a major factor in environmental pollution. The collagen is used as a structural protein, has very rich content in fish processing byproducts, and can be fully utilized to produce active peptide with high added value, thereby changing waste into valuable and reducing environmental pollution to a certain extent. A large number of studies show that compared with the action of antioxidase in human body, the protein hydrolysate has higher antioxidant activity, smaller molecular weight, simple structure and easier absorption, and has double effects of improving the antioxidant capacity of organism and supplementing nutrition when being taken into the body. The collagen antioxidant active peptide prepared from the silver carp scales can reduce environmental pollution to a certain extent, improve the market and potential value of fish processing byproducts, and has wide market prospect and positive practical significance.
The pullulan is extracellular polysaccharide secreted by aureobasidium pullulans, has the characteristics of edibility, no color or odor, no side effect on human bodies, good air tightness, plasticity, easy adhesion and the like, and is a novel multifunctional fresh-keeping material with high economic value and development potential. At present, more researchers research the application of the preservative in fruit and vegetable preservation, and the preservative is added into the preservative to be used as an edible preservative film to improve the quality of the fruit and vegetable and prolong the shelf life. However, no report has been found about the application of pullulan in the processing of medicinal plant raw materials as an edible preservative film.
The jasmine contains abundant flavone and flavonoid components, can remove free radicals, resist oxidation and inhibit food-borne pathogenic bacteria to a certain extent. The researches show that the jasmine flower extract also has the physiological functions of cancer prevention, cancer resistance, senility delay and the like. And the jasmine has light faint scent, and the adsorption and masking effects of the jasmine can be utilized to endow food with the fragrance of the jasmine, so that unpleasant smell of the jasmine is removed.
Disclosure of Invention
The invention aims to solve the defects in the prior art and provides a formula and application of a preservative and antistaling agent composition containing ferulic acid hexyl ester.
The technical scheme adopted by the invention is as follows:
the formula of the preservative and antistaling agent composition containing the ferulic acid hexyl ester comprises the following raw materials in parts by weight: 1-2 parts of hexyl ferulate, 1-2 parts of silver carp scale collagen peptide, 0.5-1 part of jasmine flower extract and 4-5 parts of pullulan.
The invention develops a formula of a preservative and antistaling agent composition containing ferulic acid hexyl ester aiming at the characteristics of fresh dendrobium officinale strips. Wherein the ferulic acid hexyl ester has the strongest bacteriostasis in a series of ferulic acid fatty acid esters with different carbon chain lengths, and can effectively inhibit the growth of food-borne pathogenic bacteria; the silver carp and fish scale collagen peptide is obtained from fish processing byproducts, has high antioxidant activity, can prevent the oxidation failure of nutrient components in the fresh dendrobium officinale strips, and can effectively reduce the phenomenon that the fresh dendrobium officinale strips are acidified due to self metabolism; the pullulan can be biodegraded, is non-toxic and tasteless, can enable the preservative and fresh-keeping agent composition to form a viscous solution, is beneficial to being attached to the surface of the fresh dendrobium officinale strips, can block gas exchange between the interior of the dendrobium officinale and the outside, weakens the respiratory strength of the dendrobium officinale, reduces water evaporation, reduces the metabolism and inhibits the occurrence of enzymatic browning; the jasmine water extract contains a large amount of flavonoids, has the effects of scavenging free radicals, resisting oxidation, protecting color and keeping freshness, can keep fresh green of dendrobium officinale fresh strips, can effectively ensure the activity of dendrobium officinale fresh strips when being prepared into solid matters, and has a certain covering effect on the herbal medicine taste of the dendrobium officinale fresh strips due to the faint scent of jasmine.
The preparation and application of the formula of the preservative and antistaling agent composition containing the ferulic acid hexyl ester comprise the following steps:
(1) preparing a jasmine flower extract: removing impurities from flos Jasmini sambac, cleaning, drying in the shade, adding into a grinder, and grinding to obtain flos Jasmini sambac powder; adding jasmine pollen powder into a distillation retort, adding 3-5 times of water for extraction for 2 hours, filtering after extraction, storing filtrate, adding the same amount of water into filter residue, extracting again and filtering; adding cellulase accounting for 0.2-0.4 percent of the weight of the jasmine powder into the filtrate collected at the two times for enzymolysis for 30min at normal temperature; after enzymolysis, introducing water vapor from the bottom of a distillation retort, continuously refluxing and heating for 1h at 95 ℃, quickly concentrating by a vacuum film device to obtain a jasmine flower extracting solution, and finally freeze-drying the jasmine flower extracting solution at-40 ℃ for 12h to obtain a solid jasmine flower extract.
(2) Dissolving and compounding: weighing hexyl ferulate, silver carp scale collagen peptide, jasmine flower extract and pullulan in sequence according to the proportion, dissolving in distilled water, adding glycerol, and mixing the components uniformly to obtain the preservative and antistaling agent composition solution
(3) Soaking, smearing, packaging and storing: processing the picked fresh dendrobium officinale strips, removing withered and shriveled parts, cleaning, soaking the cut of the fresh dendrobium officinale strips in the prepared preservative composition for 1-2 min, uniformly coating the rest preservative composition on the fresh dendrobium officinale strips, vacuum packaging and refrigerating.
Wherein the mass ratio of the hexyl ferulate to the glycerol in the preservative and antistaling agent composition solution is 2: 1; the presence of glycerol enables the hexyl ferulate to be dispersed more uniformly in the preservative and antistaling agent composition solution.
Wherein the amount of distilled water in the preservative and antistaling agent composition solution can be added according to the requirement.
Wherein the mass ratio of the dendrobium officinale fresh strips to the solid preservative and antistaling agent composition is 1: 0.03-0.06.
Wherein the refrigerating temperature of the fresh dendrobium officinale strips is 3-5 ℃.
Compared with the prior art, the invention has the following beneficial effects:
(1) the invention adopts the ferulic acid hexyl ester as one of the effective components in the solid preservative and fresh-keeping agent. Compared with unmodified ferulic acid, the ferulic acid hexyl ester has increased lipid solubility, widened application range in food, and stronger antibacterial and antioxidant activities, and can effectively inhibit the growth of pathogenic organisms in fresh dendrobium officinale strips and prolong the putrefaction time of the fresh dendrobium officinale strips.
(2) The collagen antioxidant active peptide prepared by the method effectively utilizes the fish processing by-product, namely the silver carp scales, can prevent the oxidation failure of nutrient components in the fresh dendrobium officinale strips, effectively reduces the phenomenon that the fresh dendrobium officinale strips are acidified due to self metabolism, is beneficial to the storage of the fresh dendrobium officinale strips, can reduce the environmental pollution to a certain extent, improves the market and potential value of the fish processing by-product, and has wide market prospect and positive realistic significance.
(3) The invention also prepares the solid preservative and antistaling agent composition by compounding the ferulic acid hexyl ester, the silver carp scale collagen peptide, the jasmine flower extract and the pullulan. The invention effectively solves the problems of putrefaction, color change, water loss and the like of the fresh dendrobium officinale strips in storage by using less raw materials; and the treated fresh dendrobium officinale strips not only can keep the efficacy for a long time, but also effectively reduce the herbal flavor of the fresh dendrobium officinale strips.
(4) The fresh dendrobium officinale strips produced by the method have long storage period and high nutritional value and safety, cater to the consumption concept of modern consumers, and improve the commercial value of the fresh dendrobium officinale strips.
(5) The preservative developed by the invention is solid, is more convenient to apply and transport compared with a liquid preservative, and meets the market demand.

Detailed Description
The present invention will be further described with reference to specific examples, and "parts" in the present invention are parts by weight unless otherwise specified. The method for measuring the total number of colonies in examples and comparative examples is described in GB 4789.2-2016, and the method for measuring the water loss rate in examples and comparative examples is described in the first appendix IX H (baking method) of the pharmacopoeia of the people's republic of China, 2005 edition.
Example 1
The formula of the preservative and antistaling agent composition containing the ferulic acid hexyl ester comprises the following components in parts by weight: 1 part of hexyl ferulate, 1 part of silver carp scale collagen peptide, 0.5 part of jasmine flower extract and 4 parts of pullulan.
The preparation and application of the preservative and antistaling agent composition comprise the following steps:
(1) preparing a jasmine flower extract: removing impurities from flos Jasmini sambac, cleaning, drying in the shade, adding into a grinder, and grinding to obtain flos Jasmini sambac powder; adding jasmine pollen powder into a distillation retort, adding 3-5 times of water for extraction for 2 hours, filtering after extraction, storing filtrate, adding the same amount of water into filter residue, extracting again and filtering; adding cellulase accounting for 0.2-0.4 percent of the weight of the jasmine powder into the filtrate collected at the two times for enzymolysis for 30min at normal temperature; after enzymolysis, introducing water vapor from the bottom of a distillation retort, continuously refluxing and heating for 1h at 95 ℃, quickly concentrating by a vacuum film device to obtain a jasmine flower extracting solution, and finally freeze-drying the jasmine flower extracting solution at-40 ℃ for 12h to obtain a solid jasmine flower extract.

After the fresh dendrobium officinale strips are stored for six months according to the method, the fresh dendrobium officinale strips are bright green and full, and no moldy peculiar smell is found in all the fresh dendrobium officinale strips in the same batch. The total number of colonies is less than 1.2X 103CFU/g, average water loss 0.50%.
Example 2
The formula of the preservative and antistaling agent composition containing the ferulic acid hexyl ester comprises the following components in parts by weight: 2 parts of hexyl ferulate, 2 parts of silver carp scale collagen peptide, 1 part of jasmine flower extract and 5 parts of pullulan.
The preparation and application of the preservative and antistaling agent composition comprise the following steps:

After the fresh dendrobium officinale strips are stored for six months according to the method, the fresh dendrobium officinale strips are bright green and full, and no moldy peculiar smell is found in all the fresh dendrobium officinale strips in the same batch. The total number of colonies is less than 0.8X 103CFU/g, average water loss rate of 0.32%.
Example 3
The formula of the preservative and antistaling agent composition containing the ferulic acid hexyl ester comprises the following components in parts by weight: 1.5 parts of ferulic acid hexyl ester, 1.5 parts of silver carp scale collagen peptide, 0.8 part of jasmine flower extract and 4.5 parts of pullulan.
The preparation and application of the preservative and antistaling agent composition comprise the following steps:
(1) preparing a jasmine flower extract: removing impurities from flos Jasmini sambac, cleaning, drying in the shade, adding into a grinder, and grinding to obtain flos Jasmini sambac powder; adding jasmine pollen powder into a distillation retort, adding 3-5 times of water for extraction for 2 hours, filtering after extraction, storing filtrate, adding the same amount of water into filter residue, extracting again and filtering; adding cellulase accounting for 0.2-0.4 percent of the weight of the jasmine powder into the filtrate collected at the two times for enzymolysis for 30min at normal temperature; after enzymolysis, introducing water vapor from the bottom of a distillation retort, continuously refluxing and heating for 1h at 95 ℃, quickly concentrating by a vacuum film device to obtain a jasmine flower extracting solution, and finally freeze-drying the jasmine flower extracting solution at-40 ℃ for 12h to obtain a solid jasmine flower extract.

(3) Soaking, smearing, packaging and storing: processing the picked fresh dendrobium officinale strips, removing withered and shriveled parts, cleaning, soaking the cut openings of the fresh dendrobium officinale strips in the prepared preservative composition for 2min according to the mass ratio of the fresh dendrobium officinale strips to the solid preservative composition of 1:0.05, then uniformly coating the rest preservative composition on the fresh dendrobium officinale strips, and refrigerating at 4 ℃ after vacuum packaging.

Comparative example 1
The comparison example mainly explores the storage period change of the fresh dendrobium officinale strips when the preservative composition is not coated during storage, and compared with the example 1, the difference is that the preservative composition is not coated after the fresh dendrobium officinale strips are pretreated, and the fresh dendrobium officinale strips are directly subjected to vacuum packaging and refrigeration.
After the fresh dendrobium officinale strips are stored for six months according to the method, the fresh dendrobium officinale strips are dark in color and shriveled, and about 80% of all the fresh dendrobium officinale strips in the same batch have moldy peculiar smell. The total number of colonies is more than 104CFU/g, average water loss 29.64%.
Comparative example 2
Compared with theembodiment 1, the difference is that the applied preservative composition is composed of the following components in parts by weight after the dendrobium officinale fresh strips are pretreated: 1 part of silver carp scale collagen peptide, 0.5 part of jasmine flower extract and 4 parts of pullulan, and the total solid mass of the preservative and fresh-keeping agent composition is the same as that of theembodiment 1.
After the fresh dendrobium officinale strips are stored for six months according to the method, the fresh dendrobium officinale strips are darker and unsaturated, and about 18 percent of all the fresh dendrobium officinale strips in the same batch have moldy peculiar smell. The total number of colonies is less than 6.7X 103CFU/g, average water loss 1.67%.
Comparative example 3
Compared with theembodiment 1, the difference is only that after the dendrobium officinale fresh strips are pretreated, the applied preservative is the ferulic acid hexyl ester which is dispersed by glycerol and water, and the quality of the ferulic acid hexyl ester is the same as the total solid mass of the preservative composition used in theembodiment 1.
After the fresh dendrobium officinale strips are stored for six months according to the method, the fresh dendrobium officinale strips are darker and unsaturated, and about 5 percent of all the fresh dendrobium officinale strips in the same batch have moldy peculiar smell. The total number of colonies is less than 4.9X 103CFU/g, average water loss 20.84%.
After the preservative composition obtained in theembodiment 1 and the comparative examples 1-3 is applied to fresh dendrobium officinale strips, the total number of bacterial colonies and the time change curve are shown in figure 1, and the water loss rate and the time change curve are shown in figure 2.
