PANO1 is a proapoptotic protein that posttranslationally modulates expression of the tumor suppressor p14ARF (600160) (Watari et al., 2012).
By screening for tumor-related genes,Watari et al. (2012) isolated the cDNA for human PANO1, which they called PANO. The putative protein contains 215 amino acids and has a calculated molecular mass of 23.7 kD. The C-terminal half of PANO has a proline-rich region flanked by 2 arginine-rich regions. RT-PCR analysis showed that PANO was ubiquitously expressed in human tissues. Immunofluorescence assays revealed predominant localization of HA-tagged PANO protein to the granular component region of nucleolus in transfected Hela cells. The C-terminal arginine-rich region of PANO was essential for nucleolar localization.
Watari et al. (2012) found that ectopic expression of PANO induced apoptosis in HeLa cells in a low-serum medium. The C-terminal region of PANO was required for its proapoptotic function. PANO colocalized with p14ARF in nucleolus, and PANO expression increased the expression level of p14ARF by stabilizing the p14ARF protein. Knockdown analysis in Hela cells confirmed that PANO was necessary for stable expression of p14ARF protein and showed that PANO-induced apoptosis was mediated by p14ARF. PANO also suppressed tumorigenicity in vivo, as PANO-expressing Hela cell clones did not form tumors in nude mice.
