I was thrilled by this milestone paper because it brings human interactome mapping into the realm of high precision, which is essential to make it a successful resource for functional genomics and systems biology studies.This is achieved by implementation of a suite of well-devised and highly stringent yeast-two-hybrid screens followed by downstream co-affinity purification validation assays.Similarly, all conclusions about sensitivity and specificity of the detected interactions are backed up by precise statistical power analyses. As a result, the deciphered physical scaffold of human protein interactions, although covering a small fraction of the global interactome space, is a highly reliable source of functional hypotheses with relevance to human physiology and disease. Importantly, since this resource is based on the Mammalian Gene Collection (MGC) of full-coding cDNAs, it can only expand in scope by testing additional clones with the standardized assays used, and benefit from integration of protein and mRNA expression profiling established with MGC clones.
