Alestramustine (INNTooltip International Nonproprietary Name), also known asestradiol 3-(bis(2-chloroethyl)carbamate) 17β-(L-alaninate), is acytostaticantineoplastic agent which was never marketed.[1][2] It is theL-alanineester ofestramustine, which is a combination of thenitrogen mustardnormustine coupled via acarbamate to theestrogenestradiol.[1][3] Alestramustine acts as aprodrug to estramustine, and also forms estradiol as abyproduct.[1][3] The drug, via itsactive metabolites, binds tomicrotubule-associated proteins andβ-tubulin and interferes withmicrotubule function, thereby inhibitingcell division.[1][3] Due to its estrogenmoiety, alestramustine is selectively concentrated inestrogen receptor-positivecells such asprostate andbreast.[1]
