The precise and uniform duplication of a cell's genome is essential to its normal growth. Focal points for the orchestration of this central process are provided by specific loci within the genome that function as sites of initiation of DNA replication. In this paper we present results that allow us to address the structural characteristics of DNA replication origins in yeast.

Initiation of DNA replication in the yeastSaccharomyces occurs at multiple sites in the genome. The average distance between initiation sites in yeast DNA isolated from S-phase cells has been determined by fiber autoradiography to be approximately 100 kb (Petes and Williamson 1975a; Rivin and Fangman 1980). More recently, a better measure of the average interorigin distance has been obtained by electron microscopic analysis of DNA isolated from cells synchronously entering S phase (Newlon and Burke 1980).
