Gen CASP14 kodira člana porodicecistein-aspartanihproteaza (kaspaza). Sekvencna aktivacija kaspaza ima centralnu ulogu u fazi izvršenjaćelijske apoptoze. Kaspaze postoje kao neaktivniproenzimi koji se podvrgavajuproteolitskoj obradi na konzerviranim asparaginskim ostacima kako bi se dobile dvije podjedinice, veliku i malu, koje se dimeriziraju i formiraju aktivni enzim. Pokazalo se da je ova kaspaza obrađena i aktivirana pomoćukaspaze-8 i10in vitro, i anti-Fas agonističkogantitijela ili TNF-ove apoptoze koja indukujeligandin vivo. Ekspresija i obrada ove kaspaze može biti uključena ukeratinocitnu terminalnu diferencijaciju, što je važno za formiranje kožne barijere.[7]
