Is AM404 save to use? Is it toxic or nontoxic for the liver? Paracetamol is toxic for the liver. Maybe they should sell AM404 instead of the paracetamol :)

just curious what other people think, but couldn't am404 also prevent inflammation by preventing the synthesis of prostaglandins, since they are derived from arachadonic acid? (Ccroberts(t ·c ·g )21:07, 15 October 2007 (UTC))[reply]

From my understanding of the subject, there exists no definite conclusions reguarding paracetamol's mechanism of action. It is believed that AM404 is responsible for much (or all) of the analgesic action. Curiously, however, it has relatively weak anti-inflammatory action in comparison to that ofaspirin and otherNSAIDS.--Metalhead94 (talk)00:54, 22 October 2008 (UTC)[reply]

TRPV1 agonist?

It makes no sense. TRPV1 activation causes pain or heat sensation. Capsaicine is known to activate the receptor, and that's what makes chili pungent. If one of the reasons for AM404 being an analgetic was TRPV1 activation, it would have to be the opposite to that of capsaicine. The only wat in which TRPV1 agonism would decrease pain is that a prolonged exposure to high doses of the agonist decreases the sensitivity, which is the case in any type of receptor activation, but this would mean that AM404 is no more an analgetic than capsaicine or any other source of pain... and the relief would come after a period of pain actually getting worse.

AM404 inhibits the uptake and degradation of arachidonyl ethanolamide,i.e., anandamide, an endogenous ligand ("endocannabinoid") at cannabinoid CB1 receptors. Blocking degradation enhances and prolongs anandamide concentrations, i.e., actions, where it is released. The pain relieving effects of acetaminophen are at least partly reduced by CB1 antagonists.Inhibition of COX-2 also enhances endocannabinoid actions. Читал в Русской Википедии,что Парацетамол - нейротоксин, после курса лечения парацетамолом людей, статистически длстоверно выявлено снижение эмоционального отклика сопереживания, при просмотре фильмов, людьми принимавшими парацетамол. Измеряли(помоему по ЭЭГ или ЯМР Томографией мозга, обширным гомогенизированным тестовым видео контентом, ИМХО могут выступать ютуб видео сборники смешного поведения животных,сборники ДТП, ЧП, ЧС и тп, предоставляющие материал для просмотра, со сравнительно равномерной плотностью эмоциональной составляющей контента) интенсивность эмоциональных реакций в ответ на значимые сюжетные кластеры, до и после лечения. Потеря(значительное ослабление) зрителем вовлечёности, и эмоциональной окраски фильмов, реакции сопереживания и тп. оказалась пожизненной... неизлечимой.
