The R25 begins in the suburb ofSaxonwold, just north of theJohannesburg Zoo, at an intersection withJan Smuts Avenue,Johannesburg'sM27 road. It heads eastwards, meeting Johannesburg'sM9 road (Oxford Road) and meeting Johannesburg'sM1 freeway (De Villiers Graaff Motorway; southbound access only), up to the junction withLouis Botha Avenue, Johannesburg'sM11 road, where it joins the road northwards up to the next junction, where it continues by a right turn as Hathorn Avenue. It then becomes George Avenue and makes its way north-east through the suburbs ofSydenham,Fairmount andSandringham, becoming Modderfontein Road and crossing theJukskei River, bypassing Edenvale Hospital, before crossing theN3 highway (Johannesburg Eastern Bypass) as a dual carriageway and passing byGreenstone Hill, where Greenstone Shopping Mall is located.[1]

It continues north-east as Modderfontein Road, with Greenstone Shopping Mall on the southern side and Longmeadow Business Estate on the northern side. After meeting theM37 municipal route fromEdenvale and theM54 municipal route fromAlexandra in Greenstone, the R25 bypassesModderfontein to the south and crosses the border between theCity of Johannesburg Metropolitan Municipality and theCity of Ekurhuleni Metropolitan Municipality. It then goes through the western and northern suburbs of the city ofKempton Park.[1]

In Kempton Park, it first passes by the area separatingKempton Park West fromEsther Park, where it crosses theM39 municipal route (Zuurfontein Avenue), the road betweenIsando in the south andMidrand/Thembisa in the north. It then becomes the road separatingEdleen andVan Riebeeck Park fromTerenure. It then becomes the road separatingBirchleigh fromNorkem Park andBirchleigh North. It proceeds to reach a t-junction with theM57 municipal route, just north of the M57's junction with theR23 route fromBenoni.[1]

The R25 joins the M57 northwards for 800 metres, bordering Esselen Park, up to the next junction (a 4-way junction) nearThembisa, where the R25 becomes the road to the right (east). Right after, it crosses theR21 highway betweenPretoria andJohannesburg International Airport and makes a direct line forBapsfontein. The distance is 20 km between Kempton Park and Bapsfontein.[1]

At Bapsfontein, the R25 meets theR50 fromPretoria and joins it going south-east for 1.6 km up until the junction with theR51 fromDaveyton (the main four-way junction of Bapsfontein). At that junction, as the R51 is the road south-west to Daveyton, the R25 becomes the same road heading north-east from that junction towards Bronkhorstspruit while the R50 continues south-east toDelmas. While pointing towards Bronkhorstspruit, the R25 meets the eastern termini of theM30 andM6municipal routes, which are coming fromGarsfontein andLynnwood inPretoria East respectively, as well as the southern termini of theR515 and theR568.[1]

The R25 meets the north-eastern terminus of theR42 just north of theBronkhorstspruit Dam right before turning north and enteringBronkhorstspruit in theCity of Tshwane Metropolitan Municipality. It intersects with theN4 highway (Maputo Corridor) and theR104 road (both coming fromPretoria in the west) and goes northwards through Bronkhorstspruit. The distance is 40 km between Bapsfontein and Bronkhorstspruit.[1]

Afterwards, the R25 temporarily goes intoMpumalanga, where it bypassesWolvenkop and intersects with theR544 fromWitbank in the town of Verena, before proceeding intoLimpopo province, where it passes throughDennilton before ending in the vicinity of the small farming town ofGroblersdal, terminating at a junction with theN11 and theR33. The distance is 110 km between Bronkhorstspruit and Groblersdal.[1]
