Large tumor suppressor kinase 1 (LATS1) is anenzyme that in humans is encoded by theLATS1gene.[5][6][7]

It has been associated with theHippo signaling pathway, where it phosphorylates YAP andTAZ to inactivate their function.[8]

The protein encoded by this gene is a putativeserine/threonine kinase that localizes to themitotic apparatus and complexes withcell cycle controllerCDC2 kinase in earlymitosis. The protein isphosphorylated in a cell-cycle dependent manner, with lateprophase phosphorylation remaining throughmetaphase. TheN-terminal region of the protein binds CDC2 to form a complex showing reducedhistone H1 kinase activity, indicating a role as a negative regulator of CDC2/cyclin A. In addition, theC-terminal kinase domain binds to its own N-terminal region, suggesting potentialnegative regulation through interference with complex formation via intramolecular binding. Biochemical and genetic data suggest a role as atumor suppressor. This is supported by studies in knockout mice showing development ofsoft-tissue sarcomas,ovarian stromal cell tumors and a high sensitivity tocarcinogenic treatments.[7]
