CYB004, orCYB-004, also known asdeuterated dimethyltryptamine (dDMT), is aserotonergic psychedelic related todimethyltryptamine (DMT) which is under development for the treatment ofgeneralized anxiety disorder.[1][4][2][3][5] It is administered byinhalation orintravenous injection.[1][2]

It is atryptaminederivative and is adeuteratedanalogue and form of DMT.[1][4][3] Thepharmacodynamic profile of CYB004, including its interactions withserotonin receptors and its effects in animals, is similar to that of DMT.[3] As with DMT, CYB004 is apotentagonist of theserotonin5-HT2A receptor and producespsychedelic-like effects in animals.[1][6][3] However, CYB004, due to its deuteration, is more resistant tometabolism than DMT and shows a longerelimination half-life (by 2.5- to 2.9-fold) and slowerclearance (by 38 to 55%) in animals.[3] Thebrain toplasma ratio of CYB004 was also increased (by 30%) relative to DMT, indicating slightly greatercentralpermeability as well.[3]

As of August 2024, CYB004 is inphase 2clinical trials for generalized anxiety disorder.[1][4] It was also under development for the treatment ofsubstance-related disorders and of otherpsychiatric disorders, but development for these indications was discontinued.[1] The drug is under development byCybin.[1][4] The exactchemical structure of CYB004 (i.e., whichhydrogenatoms are deuterated) does not yet seem to have been disclosed.[1] However, Cybinpatented deuterated tryptamines, including deuterated forms of DMT like DMT-d10, in 2023.[7] Other related drugs include the deuterated tryptamineCYB003 and the deuteratedphenethylaminesCYB005 andCYB210010.[6][2]
