Aleph, orAleph-1, also known asDOT orpara-DOT or as4-methylthio-2,5-dimethoxyamphetamine, is apsychedelic drug of theamphetamine andDOx families.[1]

Effects

Aleph was first synthesized byAlexander Shulgin, who named it after thefirst letter of theHebrew alphabet. In his bookPiHKAL, Shulgin lists the dosage range as 5 to 10 mg, with effects typically lasting for 6 to 8 hours.[1]

Pharmacology

Like many other psychedelics, Aleph is apartial agonist at the5-HT2A receptor (EC50 = 10 nM).[2][3] It has weakMAO-A inhibitory activity with anIC50 of 5.2 μM. For reference,amphetamine has an IC50 of 11 μM and4-methylthioamphetamine has a value of 0.2 μM.[4]
