The 2016 Premier League Knockout Cup was the 49th edition of theKnockout Cup for tier two teams. It was the last time it would be known as the Premier League Knockout Cup because the following season it would be theSGB Championship Knockout Cup.

Glasgow Tigers were the winners of the competition.[9]
